The beginning of one of the most sensational comics out today starts off with Norman Osborne explaining to his assistant, Justin, how Greek mythology explains the origin of the first spider. We start to see Norman's rage as a business man. He is telling someone about this new wonder drug called OZ which is going to revolutionize the world. The testing has begun on mammals and insects, and the results from spider testing have been extraordinary. While Norman is talking away his assistant throws the spider that Norman passed to him into the sample jar without closing the lid and it escapes.
We are introduced to the character of Peter Parker and we learn of all the troubles that a boy like him has in high school. Peter is quietly reading as Flash Thompson and Kong are throwing food at him and celebrating in there "Victory". Peter just ignores this and looks over to the girl of his dreams, Mary Jane Watson. As the food throwing continues Uncle Ben steps in and begins to talk to Peter and Mary Jane.
At school, Peter gets the worst of the bullying, but his good friend, Harry Osborn, steps in to help his friend Pete. Returning home Peter, Ben, and the lovable Aunt May are discussing Peters social life and how being so smart can be a burden on such a young boy. Peter just shrugs it off and continues thinking. Peter and Harry are now talking about Pete's life, his parents, and his crush on Mary Jane Watson. Harry returns home and finds his father talking on the phone with someone, while totally ignoring Harry considering him more a pest then his son.
The book then goes into the most integral part of the story, when the class takes a trip to Osborne Industries where the super drug OZ is being tested. Out of nowhere Peter feels a great pain as a enormous spider bites his hand. Peter flicks the spider which lands on Mary Jane and falls to the floor. He blacks out and wakes up only to get taunted by his classmates again for being such a wimp. Norman Osborne becomes informed of this incident and instead of worrying about it says that he wants to keep a eye on Peter.
At school, Kong tries to kick Peter in the backside, but something tells Peter this is coming so he turns and flips Kong over. Everyone is in shock and Peter collapses to the floor again. He wakes up in the hospital for blood tests, but someone steals Peter’s blood sample to bring back to Oscorp. Judging by the results of the blood test they believe Peter will eventually die.
Back at the Parker home, Aunt May tries to help Peter overcome his "allergy" to the spider bite. As Peter walks to school, someone tries to run him over with their car. Peter does a humongous back-flip over the car and begins to realize that something is wrong with him. Peter returns home to try and figure out what is happening to him. He starts to do some research on spiders and his own blood. Peter skipped all day of school for his research so Aunt May and Uncle Ben start scolding him for skipping class and send him to his room. As Peter is thinking about what happened today he stares up at his ceiling and almost like a instinct he climbs up the wall.
